光声(PA)成像作为一种结合了光学和声学成像优势的新型成像方式,具有深层组织穿透和高空间分辨率等优点,在重大疾病的早期影像诊断方面有着巨大的应用前景。然而传统的PA造影剂依然存在信噪比低、选择性及特异性差等不足,容易产生假阳性诊断结果。激活型PA造影剂可以有效的降低背景噪声,并提升成像的灵敏度和特异性,是目前PA造影剂设计与构筑的主要趋势。本综述首先简单介绍了PA成像的原理,然后结合近几年在金属离子、酶、活性氮和活性氧等相关方面的生物成像应用,梳理了可激活探针在不同微环境中的响应方式。最后,对激活型探针在PA成像中的应用进行了总结和展望。  相似文献

Gliomas are the most common malignant brain tumors in adults, characterized by a high proliferation and invasion. The tumor microenvironment is rich in growth-promoting signals and immunomodulatory pathways, which increase the tumor’s aggressiveness. In response to hypoxia and glioma therapy, the amounts of adenosine triphosphate (ATP) and adenosine diphosphate (ADP) strongly increase in the extracellular space, and the purinergic signaling is triggered by nucleotides’ interaction in P2 receptors. Several cell types are present in the tumor microenvironment and can facilitate tumor growth. In fact, tumor cells can activate platelets by the ADP-P2Y12 engagement, which plays an essential role in the cancer context, protecting tumors from the immune attack and providing molecules that contribute to the growth and maintenance of a rich environment to sustain the protumor cycle. Besides platelets, the P2Y12 receptor is expressed by some tumors, such as renal carcinoma, colon carcinoma, and gliomas, being related to tumor progression. In this context, this review aims to depict the glioma microenvironment, focusing on the relationship between platelets and tumor malignancy.  相似文献

以顺铂为代表的小分子铂类抗癌药物是临床应用的一线化疗药物,但其严重的毒副作用和难以克服的耐药性限制了铂类药物的临床应用和研发。运用纳米药物递送技术可以实现药物的靶向递送和可控释放,来提高药物的生物利用度,降低药物的毒副作用以及耐药性,为癌症的治疗带来新的希望。此外,丰富多样的纳米递送体系易于实现药物与具有生物学活性试剂的共运输,从而为各种治疗策略以及诊疗策略的联用提供可能,为最终实现癌症的精准治疗展现广阔前景。本文从靶向递药、药物可控释放、联合治疗、诊疗一体化四个方面对铂类抗癌药物的多功能纳米递送体系在癌症治疗中的最新研究进展进行综述,同时通过列举最新研究成果,展示了新材料、新技术以及新颖设计思想在铂基纳米递送体系中的应用。  相似文献

Through the phase inversion technique, asymmetric flat sheet pH-responsive Polysulfone (PSF) membrane was prepared and utilized for recovering H2SO4 in the presence of NaCl and KHCO3 from wastewater. Hydrophilic and pH-responsive characteristics were incorporated within the membrane by blending Polyethylene glycol methyl ether (PEGME) and Humic acid (HA). The modification in membrane morphology with pH was characterized by Field Emission Scanning Electron Microscopy (FESEM), Differential scanning calorimetry (DSC) and Fourier Transform Infrared Studies (FTIR) method. The ion exchange capacity of the prepared pH-responsive membrane increased from 0.145 to 0.25 mmol/g when compared to the pristine PSF membrane. Pure water flux (PWF) of 113.8–46.8 L/m2h, water uptake of 25.9%–6.8% were obtained for pH-responsive membrane when pH varied from 4 to 12. Recovery of H2SO4 was optimized by design expert software 9.0 TRIAL and was found to be a maximum of 76.57 ± 1.5% in the presence of 0.32 M NaCl and 0.5 M KHCO3 at pH ~8.4, through the pH-responsive PSF membrane by diffusion dialysis process. The influencing parameters (pH, NaCl (M) and KHCO3 (M)) were optimized and acid recovery modeling was performed through response surface methodology (RSM) and central composite design (CCD). F value of 6573.40 through ANOVA study indicated the significance of the quadratic model chosen, whereas an insignificant lack of fit (prob > F = 0.0519) confirmed the goodness of fit between the model and obtained experimental data's.  相似文献

User-friendly, low-cost equipment for preventive screening of severe or deadly pathologies are one of the most sought devices by the National Health Services, as they allow early disease detection and treatment, often avoiding its degeneration. In recent years more and more research groups are developing devices aimed at these goals employing gas sensors. Here, nanostructured chemoresistive metal oxide (MOX) sensors were employed in a patented prototype aimed to detect volatile organic compounds (VOCs), exhaled by blood samples collected from patients affected by colorectal cancer and from healthy subjects as a control. Four sensors, carefully selected after many years of laboratory tests on biological samples (cultured cells, human stools, human biopsies, etc.), were based here on various percentages of tin, tungsten, titanium, niobium, tantalum and vanadium oxides. Sensor voltage responses were statistically analyzed also with the receiver operating characteristic (ROC) curves, that allowed the identification of the cut-off discriminating between healthy and tumor affected subjects for each sensor, leading to an estimate of sensitivity and specificity parameters. ROC analysis demonstrated that sensors employing tin and titanium oxides decorated with gold nanoparticles gave sensitivities up to 80% yet with a specificity of 70%.  相似文献

A series of physiological barriers have impeded nanoparticle-based drug formulations (NDFs) from reaching their targeted sites and achieving therapeutic outcomes. In this study, we develop size-controllable stealth doxorubicin-loaded nanodrug coated with CD47 peptides (DOX/sNDF-CD47) based on supramolecular chemistry to overcome multiple biological barriers. The smart DOX/sNDF-CD47 can efficiently decrease sequestration by macrophages and disassemble into poly(amidoamine) dendrimers with nuclear localization sequences (DOX/PAMAM-NLS) in the presence of matrix metalloproteinase-2 (MMP-2). Such structure transformation endows DOX/sNDF-CD47 with the ability of deep penetration in multicellular tumor spheroid, lysosomal escape, and nucleus localization, resulting in excellent cytotoxicity and drug resistance combating. In vivo experiments further confirmed that DOX/sNDF-CD47 has good tumor-targeting ability and can significantly improve therapeutic efficacy of DOX on xenograft tumor model. The ability to overcome multiple biological barriers makes sNDF-CD47 a promising NDFs to treat cancer expressing MMP-2 and combating drug resistance.  相似文献
